Abstract
Over the course of the past 50 years, lipid emulsions have proven to be an effective answer to clinical as well as formulation-derived challenges. Parenterally administered lipid emulsions are a well-established source of calories, essential fatty acids, and vitamins for hospitalized patients in need for parenteral nutrition. Their natural and well tolerated excipients pose a major advantage regarding undesired adverse effects upon injection and can be administered even for prolonged durations without health complications. Due to the incorporated oil phase, they also pose a formidable option for solubilization of highly lipophilic drug molecules that cannot be formulated via solubilization in aqueous media. This becomes more and more relevant, as novel developed drug substances tend to be small molecules with highly lipophilic properties.
The right choice of pharmaceutically approved excipients that fit the purpose of the respective lipid emulsion and are compatible with formulation, packaging material and production process, poses an ongoing challenge for formulation experts in the pharmaceutical industry. This goes hand in hand with the development of validated and pharmaceutically compliant production processes that produce resilient lipid emulsions with a prolonged shelf-life.
This seminar aims to give an overview of the most important critical process parameters of lipid emulsion production as well as critical quality aspects of the finished product that are required by regulatory guidelines and authorities.
