Abstract

After a short introduction presenting the most recent examples in high resolution cryo electron microscopy (EM) of membrane proteins, this seminar will rather focus on morphological EM studies with self-assembled soft matter aggregates. Different preparation techniques for EM samples, as well as advantages, disadvantages, and artefacts of negative stain and cryo EM will be discussed considering the following questions:

  • Negative stain or cryo EM: Is cryo always better?
  • Structural pathways of vesicle solubilization: Is there a limit?
  • Visualization of nanodiscs: What can we learn from negative stain and cryo EM?
